目的 探讨Ras相关的C3肉毒素底物1(Rac1)基因在子宫内膜癌组织中表达情况及其临床意义。
方法 应用实时荧光定量PCR检测96例子宫内膜癌和40例正常子宫内膜组织Rac1基因的表达,并分析其与子宫内膜癌病人生存的关系以及子宫内膜癌病人预后的影响因素。
结果 子宫内膜癌组织Rac1 mRNA相对表达量为1.75±0.16,显著高于正常子宫内膜组织(1.38±0.14),差异有统计学意义(t=14.095,P<0.01)。子宫内膜癌组织Rac1 mRNA相对表达量与肿瘤分化程度、浸润深度、淋巴结转移和血管浸润有关(t=5.322~9.739,P<0.05)。Kaplan-Meier生存分析显示,Rac1低表达组病人平均生存时间为58.82个月,高表达组为45.98个月,两组比较差异有显著性(χ2=4.972,P<0.05)。Cox比例风险回归模型分析显示,肿瘤分化程度、淋巴结转移和Rac1 mRNA相对表达量是影响子宫内膜癌病人预后的风险因素(HR=2.449~13.582,P<0.05)。
结论 Rac1基因在子宫内膜癌组织中呈高表达,是影响预后的风险因素之一。
Abstract:
Objective  To investigate the expression of Ras-related C3 botulinum toxin substrate 1 (Rac1) gene in endometrial carcinoma tissue and its clinical significance.
Methods  Quantitative real-time PCR was used to measure the expression of the Rac1 gene in 96 cases of endometrial carcinoma and 40 cases of normal endometrium tissues. The relationship between the expression of the Rac1 gene and the survival of patients with endometrial carcinoma, as well as the prognostic factors, was then analyzed.
Results  The relative expression level of Rac1 mRNA in endometrial carcinoma tissues was 1.75±0.16, which was significantly higher than that in the normal endometrial tissues (1.38±0.14) (t=14.095,P<0.01). The relative expression level of Rac1 mRNA in endometrial carcinoma tissues was related to the degree of tumor differentiation, depth of invasion, lymph node metastasis, and blood vessel invasion (t=5.322-9.739,P<0.05). Kaplan-Meier survival analysis showed that the low Rac1 expression group had a significantly longer mean survival time than the high Rac1 expression group (58.82 vs 45.98 months,χ2=4.972,P<0.05). Cox proportional-hazards regression model analysis showed that tumor differentiation degree, lymph node metastasis, and the relative expression level of Rac1 mRNA were prognostic factors in patients with endometrial carcinoma (hazard ratio(HR)=2.449-13.582,P<0.05).
Conclusion  High expression of Rac1 in endometrial carcinoma tissue is one of the risk factors for prognosis.
