摘要:
目的 探讨低分子肝素(LMWH)联合5-氟尿嘧啶(5-FU)对荷H22肝癌小鼠血液中组织因子(TF)及组织因子途径抑制物(TFPI)浓度的影响。
方法 选择40只昆明小鼠,随机分为空白对照组、荷瘤组、LMWH组、LMWH联合5-FU组(联合组)和5-FU组,各8例。制备H22肝癌荷瘤小鼠模型(空白对照组除外),各组分别予以生理盐水、生理盐水、LMWH、LMWH+5-FU、5-FU治疗2周。治疗结束后,经心脏取血,应用ELISA法检测各组血液中TF、TFPI的浓度,并进行比较。
结果 荷瘤组小鼠血液TF浓度高于空白对照组,TFPI浓度低于空白对照组;LMWH组、联合组及5-FU组小鼠血液中TF的浓度均低于荷瘤组,TFPI的浓度高于荷瘤组,差异有显著性(F=82.534、34.755,P<0.05)。LMWH组、5-FU组、联合组小鼠血液中TF、TFPI浓度比较,差异无统计学意义(P>0.05)。
结论 LMWH与5-FU均对肝癌小鼠TF有抑制作用,两者都可提高TFPI含量,但二者无协同作用。
Abstract:
Objective  To investigate the effect of low-molecular-weight heparin (LMWH) and 5-fluorouracil (5-FU) on the blood levels of tissue factor (TF) and tissue factor pathway inhibitor (TFPI) in mice with H22 liver cancer.
Methods  A total of 40 Kunming mice were randomly divided into blank group, tumor-bearing group, LMWH group, LMWH+5-FU group (combination group), and 5-FU group, with 8 mice in each group. All mice except those in the blank group were used to establish a model of H22 liver cancer, and the five groups were treated with normal saline, normal saline, LMWH, LMWH+5-FU, and 5-FU, respectively. Blood samples were collected from the heart after treatment, and ELISA was used to measure the blood levels of TF and TFPI.
Results  Compared with the blank group, the tumor-bearing group had a significantly higher level of TF and a significantly lower level of TFPI; compared with the tumor-bearing group, the LMWH group, the combination group, and the 5-FU group had a significantly lower level of TF and a significantly higher level of TFPI (F=82.534 and 34.755,P<0.05). There were no significant differences in the blood levels of TF and TFPI between the LMWH group, the 5-FU group, and the combination group (P>0.05).
Conclusion  Both LMWH and 5-FU have an inhibitory effect on TF and can increase the level of TFPI, but there is no superposition between them.
