摘要:
目的 探讨低频和高频有氧运动对老年大鼠骨骼肌的影响。
方法 选择健康雄性的Wistar大鼠20只,其中青年大鼠(A组)5只,老年大鼠15只,随机将15只老年大鼠分为空白对照组(B组)5只、低频有氧运动组(C组)5只、高频有氧运动组(D组)5只。A组和B组大鼠不做任何处理,C组和D组大鼠分别行低频、高频有氧运动,共8周。第8周运动结束后立刻测量各组大鼠腓肠肌处皮温,并处死大鼠测定腓肠肌质量、血管内皮生长因子(VEGF)与胶质细胞源性神经生长因子(GDNF)表达,同时观察大鼠腓肠肌形态变化情况。
结果 C组和D组大鼠腓肠肌肌纤维明显增粗,间隙变小,形态较A、B组规则;4组VEGF表达组间比较差异有显著性(F=18.37,P<0.05),各组两两间比较差异均有统计学意义(P<0.05);4组腓肠肌内GDNF表达组间比较差异有显著性(F=15.03,P<0.05),除C、D组间比较差异无显著性外(P>0.05),其余各组两两间比较差异均有统计学意义(P<0.05);4组腓肠肌质量组间比较差异有显著性(F=33.46,P<0.05),除C、D组间比较差异无显著性外(P>0.05),其余各组两两间比较差异均有统计学意义(P<0.05);4组腓肠肌/大鼠体质量比值组间比较差异有显著性(F=128.34,P<0.05),除C、D组间比较差异无显著性外(P>0.05),其余各组两两间比较差异均有统计学意义(P<0.05)。
结论 低频和高频有氧运动可显著提高老年大鼠的腓肠肌质量,并提高大鼠腓肠肌组织内VEGF和GDNF的表达,延缓老年大鼠腓肠肌的衰老性改变。
Abstract:
Objective  To investigate the effect of low-and high-frequency aerobic exercise on skeletal muscle in aged rats.
Methods  A total of 20 healthy male Wistar rats were selected, among which there were 5 young and middle-aged rats (group A) and 15 aged rats. The 15 aged rats were randomly divided into blank control group (group B), low-frequency aerobic exercise group (group C), and high-frequency aerobic exercise group (group D), with 5 rats in each group. The rats in groups A and B were not given any treatment, and those in groups C and D were given low-and high-frequency aerobic exercise, respectively; the course of treatment was 8 weeks. Skin temperature of the gastrocnemius was measured immediately after exercise at week 8, and the weight of the gastrocnemius and the expression of vascular endothelial growth factor (VEGF) and glial cell line-derived neurotrophic factor (GDNF) were measured after the rats were sacrificed. The morphological changes of the gastrocnemius were also observed.
Results  Compared with groups A and B, groups C and D had thicker muscle fibers of the gastrocnemius and smaller intermuscular space. There was a significant difference in the expression of VEGF between the four groups (F=18.37,P<0.05), and there was also a significant difference between any two groups (P<0.05). There was a significant difference in the expression of GDNF between the four groups (F=15.03,P<0.05), and there was a significant difference between any two groups except between groups C and D (P<0.05). There was a significant difference in the weight of the gastrocnemius between the four groups (F=33.46,P<0.05), and there was a significant difference between any two groups except between groups C and D (P<0.05). There was a significant difference in gastrocnemius/body weight ratio between the four groups (F=128.34,P<0.05), and there was a significant difference between any two groups except between groups C and D (P<0.05).
Conclusion  Both low- and high-frequency exercise can significantly improve the weight of the gastrocnemius, increase the expression of VEGF and GDNF in the gastrocnemius, and delay the aging changes of the gastrocnemius in aged rats.
